Abstract:

BARBUS SHARPEYI in family Cyprinidae is an endemic fish of the province Khuzestan of Iran. These fish are produced by artificial breeding using carp pituitary extract (CPE). According to the problem of artificial breeding about this fish in Iran, the objective of this study was to obtain the effectiveness new method (LHRH-a2 hormone Combined with carp pituitary extract (3 injections) on reproduction index in BARBUS SHARPEYI. The results showed that the LHRH-a2 hormone combined with carp pituitary extract in three injection (specially treat 2) lead to high spawning success (62% average) and weight of stripped egg mass/ weight of stripped egg mass (8.49% average) and fertilization success (76.57% average). Therefore, it can be concluded that like many other cyprinids, LHRH-a2 hormone combined with carp pituitary extract (3 injection) can be effectiveness for spawning induction in BARBUS SHARPEYI.

Abstract:

The present research aimed to determine acute toxicity and evaluate the effect of sub-lethal concentrations of diazinon on some haematological and immunological parameters ofBARBUS SHARPEYIby the Organization for Economic Cooperation and Development methods. Juvenile B. SHARPEYIwere exposed to 0, 5%, 10% and 25% 96h LC50 concentrations of diazinon for 21 days. Blood samples were taken after 7, 14 and 21 days of exposure to diazinon.Haematological and biochemical parameters including: RBC, Hb, PCV and WBC, as well as serum enzymes were evaluated. Immunological indices and resistance against bacterial infection, were determined. Results showed that 96h LC50 of diazinon inB. SHARPEYI was estimated at 3.987 mg/L. Diazinon toxicity at a level of 1 and 0.4 mg/L decreased RBC, Hb, and Hematocrite, in almost all sampling periods compared. WBC and globular index decreased significantly in fish exposed to 1 mg/L diazinon on days 14 and 21 (p<0.05). Dose dependent increase in serum enzymes were seen in fish exposed to diazinon. Diazinon toxicity showed no effect on serum LDH level (p>0.05). Dose dependent decrease in serum total protein and globulin were indicated in diazinon exposed fish. Serum lysozyme and bactericidal activity decreased in T3 and T4 at days 14 and 21. Mortality following challenge toA.hydrophila increased in fish exposed to 1 mg/l diazinon. It can be concluded that diazinon is toxic toB.SHARPEYI. Therefore, the strict biosecurity should be taken into consideration when this pesticide is used in agricultural fields surrounding freshwater sources of fish cultivation.

IN THE LAST DECADES, INTEREST FOR USING FROM HERBAL MEDICATIONS AS GROWTH PROMOTERS AND IMMUNOSTIMULANTS IN FISH FARMS HAS BEEN INCREASED. AMONG IMMUNOSTIMULANTS AND IMMUNOMODULATORS, HERBAL DERIVATIVES HAVE THE LOWEST SIDE EFFECTS ON ENVIRONMENT AND THEY CAN BE PROPOSED AS A GOOD SUBSTITUTE FOR CHEMICAL DRUGS AND SUPPLEMENTS IN AQUACULTURE.

Abstract:

BACKGROUND: Benni of cyprinidae family is important for nutrition and economic and commercial species in the south of Iran and the Persian Gulf region.OBJECTVES: To investigate the importance of compatibility of fishes with different salinity in different areas and ability of osmoregulation along with maintenance of homeostasis.METHODS: For this study, 144 healthyBARBUS SHARPEYI with an average weight of 350±2.36 grams and length 25±1.25 cm in five groups were studied. The first group as control was located in municipal dechlorinated water and the next four groups respectively were kept in salinity 4ppt, 8ppt, 12 ppt and 16ppt in the same condition. On days 1, 3, 7, 14, 21 and 28 the second gill arch with maximum thickness of 0.5 cm from the left were prepared and placed in Bouin’s solution. Then the standard method of paraffin sections was done and tissue sections, 5-6 micrometer thick were prepared and stained with H& E methods.RESULT: Results showed the gradual transfer of fish to water with high salinity caused obvious changes in the number and distribution of chloride cells on different days. In particular, the changes in the size of these cells in two positions, filament and Lamella were evident.CONCLUSIONS: Due to the decrease of freshwater resources, from this research it can be concluded that theBARBUS SHARPEYI can resist the salinity without tissue changing. These findings suggest thatBARBUS SHARPEYI is compatible with salinity and the concentration of 4ppt was optimum and concentrations of 8ppt and 12 ppt were tolerable.

The aim of this study was to investigate the effects of different salinity on kidney of BARBUS SHARPEYI. For this study, 144 healthy BARBUS SHARPEYI with an average weight of 350±2.36 grams and length 25±1.25 cm in five groups were studied. The first group as control located in municipal dechlorination water and the next four groups respectively were kept in salinity 4ppt, 8ppt, 12ppt and 16ppt in the same condition. On days 1, 3, 7, 14, 21 and 28 sample of kidney with maximum thickness of 0.5 cm prepare and were placed in  bouin's solution. Then the standard method of parafin sections were done and 5- 6 micrometer thick of tissue sections prepared and stained with H&E methods. Results showed the gradual transfer of fish to water with high salinity caused obvious changes as increase the number and diameter of the glomeruli  especially in high salinity but the severity was reduced at the end of the period (p<0.05). Also highest diameter and thickness of the collecting tubules were reported in fresh water at 28 days (p<0.05). Due to the decrease of freshwater resources, from this research can be concluded that the BARBUS SHARPEYI how much can resist opposite the salinity. These findings suggest that fish BARBUS SHARPEYI was friendly with salinity and ability to set vital to different salinity.

Abstract:

Understanding the embryogenesis of Bunni is a useful tool for finding the time and suitable environmental conditions for spawning, growth requirements and use of techniques to increase the growth rate and survival. The stages of embryonic development for Bunnei (BARBUS SHARPEYI) was investigated in 22oC. The reared brood stocks were spawned in captivity condition. The oocytes were spherical, brown and very adhesive. The cleavage was fast and the first division was recorded within 15-20 minutes after fertilization. The blastulation and gastrulation were investigated in 7: 40 and 13: 10 hours after fertilization with the yolk sac diameter of 1.18±0.061mm (Mean±SD), respectively. The organogenesis was started 21 hours after the fertilization, when the blastopore was closed and notochord was formed. The embryonic stages were continued by appearance the brain, the somites and the melanophores on the yolk sac. The heart was beaten and shown the blood circulation 65 hours after the fertilization. The head, tail and pectoral fins were moved frequently. The embryo reached to pre hatching and final hatching 79: 10 and 84: 10 hours after fertilization, respectively, and the embryo existed from corion with the length of about 5.29±0.121mm (Mean±SD).
